Is it possible to Auth0 to use username instead of email when we enroll the MFA using Google Authenticator?
I know it can be rename afterwards, but is it possible to use username instead of email as the default?

Unfortunately, I could not find any way to set the username instead of email as default, as you requested.
What I recommend is creating a feature request and sharing your use case with the team on the feature-request channel.
